SOP For Cleaning and Disinfection of Microbiology Laboratory
1.0 Objective
1.1 To establish a procedure for cleaning and disinfection of the Microbiology Laboratory to maintain hygienic conditions and ensure environmental control.
2.0 Scope
2.1 This SOP is applicable to cleaning and disinfection of all areas within the Microbiology Laboratory, including General Microbiology Lab, Sterility Testing Area, and MLT Section.
3.0 Responsibility
3.1 Microbiologist or above – Microbiology Laboratory:

5.1.4 Frequency
All areas of the Microbiology Laboratory shall be cleaned and disinfected twice daily (morning and evening), unless otherwise specified.
5.2 General Microbiology Laboratory
5.2.1 Remove dust from ceilings, walls, doors, window glasses, COB, incubators, grills, and external surfaces of equipment (ovens, autoclaves, refrigerators, weighing balances, pH meters, water baths, tables, etc.) using a lint-free cloth.
5.2.2 Sweep the floor using a non-shedding, lint-free dry mop.
5.2.3 Prepare two buckets:

One containing Purified Water
-
One containing diluted disinfectant solution prepared as per SOP on Preparation and Usage of Disinfectants.
5.2.4 Wipe cleaned surfaces first with cloth wetted with Purified Water and then with diluted disinfectant.
5.2.5 Clean drains, sinks, and wash basins daily with Purified Water followed by flushing with disinfectant solution.
5.2.6 Sanitize media preparation tables and washing area tables with 70% IPA.
5.2.7 Empty waste bins twice daily and clean them with water followed by disinfectant solution.
5.2.8 Remove dust from ceilings, shelves, ducts, light fixtures, and racks using a vacuum cleaner once monthly.
5.2.9 Mop floors using approved floor cleaning solution.
5.2.10 Clean stainless steel garment and shoe cabinets in the primary change area daily using a clean, dry cloth.
5.2.11 Record cleaning details in the “Cleaning and Disinfection Record of Microbiology Laboratory”.
5.2.12 Disinfectants for General Microbiology Lab

Lysol – 2.5% v/v
-
Domex – 2.5% v/v
-
Savlon – 2.5% v/v
Rotation: Weekly rotation (one disinfectant per week).
Frequency: Twice daily.
5.3 Sterility Testing Area
5.3.1 Clean garment cabinets and used garment cabinets in Change Room–1 using lint-free cloth wetted with 70% IPA.
5.3.2 Clean floors, glass surfaces, and LAF hoods using WFI and wipe with sterile lint-free mop pads.
5.3.3 Apply disinfectant to floors and spread using sterile lint-free mop pads.
5.3.4 Allow a minimum contact time of 10 minutes.
5.3.5 Remove excess disinfectant using sterile lint-free mop pads attached to a long-handled stainless steel mop.
5.3.6 Follow cleaning sequence from Sterility Testing Room towards Change Rooms.
5.3.7 Sanitize dedicated slippers daily by dipping in disinfectant (10-minute contact time), followed by rinsing in WFI, spraying with 70% IPA, and air drying.
5.3.8 Clean autoclave doors, garment cabinets, and pass boxes daily using 70% IPA.
5.3.9 Clean all stainless steel furniture, trolleys, and surfaces daily using approved disinfectant.
5.3.10 Rotate disinfectants weekly as listed below.
Disinfectants for Sterility Testing Area

Triple 100 – 1% v/v

Combatan DS – 1% v/v

Protasan DS – 1% v/v
Rotation: Weekly rotation (one disinfectant per week).
Frequency: Twice daily.
5.4 MLT Section
5.4.1 Clean used garment boxes and crossover benches daily using 70% IPA.
5.4.2 Clean floors, glass surfaces, and LAF hoods with WFI and wipe with sterile lint-free mop pads.
5.4.3 Apply disinfectant to floors and spread using sterile lint-free mop pads.
5.4.4 Maintain a minimum contact time of 10 minutes.
5.4.5 Remove excess disinfectant using sterile lint-free mop pads and stainless steel mop.
5.4.6 Sanitize dedicated slippers daily using disinfectant (10-minute contact time), rinse with WFI, spray with 70% IPA, and allow to dry.
5.4.7 Rotate disinfectants weekly as follows:

Rotation: Weekly rotation (one disinfectant per week).
Frequency: Twice daily.
5.5 Additional Disinfection
5.5.1 Use Cidex (undiluted) for spraying in testing areas once weekly.
5.6 Documentation
5.6.1 Record all cleaning and disinfection activities in Annexure–01.
